Update a Website with Changes made in Visual Studio 2012

I have website up and running. I also have source code and able to compile it in Visual Studio 2012. I made some changes in [login to view URL], but encountered issues after uploading it to web and restarting web service. I have downloaded a publish settings file from the website but I can't find how to add this profile in Visual Studio Ultimate 2012. I would need someone to guide me how to setup my visual studio to be able to make changes and be able to upload it to web. My web hosting is [login to view URL] hosting You may try guiding me with instructions, but if I can't get it to work you would need to do a logmein session and give it to me on a step by step basis.
